Measurement of pulmonary vascular permeability (PVP) is critical to study acute lung injury, sepsis, and lunch edema classification. We provided here a visualized protocol to examine PVP in a mouse model of sepsis mediated by lipopolysaccharide. The measurement includes the steps of tool preparation, mouse anesthetization, injection of albumin labeled with Evans blue dye, lung perfusion and homogenization, addition of formamide, overnight incubation, centrifuge, measurements of optical density, and data analysis. We also compared two different intravenous injection points, external jugular vein versus retro orbital regarding the measurement. Our data showed that lipopolysaccharide significantly enhanced the mouse PVP, and the data between external jugular vein and retro orbital were comparable.
